Hello everyone, we are now introducing our FCF@UNIL Newsletter. Every month, we will give you tricks, tips and advice to make sure your flow is at its best. We will bring a monthly update that will also give you information about what's to come in the facility !

On this update, we talk about the role of Global Worksheets and Normal Worksheets in DIVA and the impact of sample washing on your data.

Finally, we would like to encourage users to follow COVID protocols and respect the restriction of one user per machine.

Which Worksheet When: Global vs Normal Worksheets

Worksheets are the place within the DIVA software where users can add plots, make gates, generate statistic plots, and show the population hierarchy. While often one worksheet is enough, you can include many more to personalize to your analysis stream. There are two kinds of worksheets, Global and Normal, and understanding the difference can help to improve your analyzer experience.

Global Worksheets

Global worksheets, the default worksheet used for samples, will have the same gating strategy for all tubes. If you adjust a gate it will be applied to all samples. Global worksheets can be distinguished by their green tabs.

Normal Worksheets

Normal worksheets, mainly used for Compensation Controls, are specific to each tube. Every tube will have a separate worksheet that allows for separate gating strategies per sample tube. If you accidentally run and record samples in the Normal worksheet, it will create new tabs for new tubes, instead of applying the same uniform worksheet. Normal worksheets have white tabs.

Conclusion

It’s important to recognize which kind of worksheet you’re in while setting up a gating strategy in DIVA. Often when moving between compensation and sample tubes we forget to move back to the Global worksheet and lose time gating in a Normal worksheet when we would prefer a Global one.

Normal worksheets created can always be converted to a Global worksheet. Just copy the Analysis element that is attached to the tube and paste it into a new Global worksheet. Feel free to ask the FCF staff for assistance in how this is done.
